Cytotoxic activity in cell cultures
Gemcitabine shows significant cytotoxic effects against a variety of cultured murine and human tumour cells. Its action is phase-specific such that gemcitabine primarily kills cells that are undergoing DNA synthesis (S-phase) and, under certain circumstances, blocks the progression of cells at the junction of the G1/S phase boundary. In vitro, the cytotoxic effect of gemcitabine is dependent on both concentration and time.
Antitumoral activity in preclinical models
In animal tumour models, antitumoural activity of gemcitabine is schedule-dependent. When gemcitabine is administered daily, high mortality among the animals but minimal antitumoural activity is observed. If, however, gemcitabine is given every third or fourth day, it can be administered in non-lethal doses with substantial antitumoural activity against a broad spectrum of mouse tumours.
Mechanism of action
Cellular metabolism and mechanism of action: Gemcitabine (dFdC), which is a pyrimidine antimetabolite, is metabolised intracellularly by nucleoside kinase to the active diphosphate (dFdCDP) and triphosphate (dFdCTP) nucleosides. The cytotoxic effect of gemcitabine is due to inhibition of DNA synthesis by two mechanisms of action by dFdCDP and dFdCTP. First, dFdCDP inhibits ribonucleotide reductase, which is uniquely responsible for catalysing the reactions that produce deoxynucleoside triphosphates (dCTP) for DNA synthesis. Inhibition of this enzyme by dFdCDP reduces the concentration of deoxynucleosides in general and, in particular, dCTP. Second, dFdCTP competes with dCTP for incorporation into DNA (self-potentiation).
Likewise, a small amount of gemcitabine may also be incorporated into RNA. Thus, the reduced intracellular concentration of dCTP potentiates the incorporation of dFdCTP into DNA. DNA polymerase epsilon lacks the ability to eliminate gemcitabine and to repair the growing DNA strands. After gemcitabine is incorporated into DNA, one additional nucleotide is added to the growing DNA strands. After this addition there is essentially a complete inhibition in further DNA synthesis (masked chain termination). After incorporation into DNA, gemcitabine appears to induce the programmed cell death process known as apoptosis.

vinos de hacienda de expresión personal
El proyecto inmobiliario-vitivinícola de “Talaza Taller de Vino” conocido como “La Melonera”, se inició hace más de 7 años de la mano entre otros de Javier Suqué Mateu (Propietario de Castillo de Perelada) y José Luis Pérez, impulsor de la Denominación de Origen Priorato, con vinos de fama y culto
mundial tales como L´Ermita, Clos Dofí, Clos Mogador y Mas Martinet.
Tomando como referencia la evolución en los últimos 20 años en la nueva cocina española, que ha llevado a situar 4 chefs españoles entre los 10 mejores del mundo y a Ferrán Adriá como el numero 1 desde 2002, “La Melonera” intenta aplicar en nuestro proyecto todas estas experiencias, con una filosofía de
investigación multidisciplinar constante tanto en la viticultura, la elaboración y el ensamblaje final o coupage de nuestros vinos.
Tras ver como muchos empresarios y profesionles se acercaban a pedir consejo
tanto a José Luis Perez o Javier Suque, para iniciar un vino de expresión personal, a imagen de los grandes Chateaus bordoleses, Vega Sicilia o los grandes Brunello di Montalcino) y gran parte de ellos renunciar a llevar a cabo dicha ilusión tanto por la complejidad, las fuertes inversiones así como la dificultad de crear algo realmente diferente, y por último, al miedo de que un
simple hobby y pasión, como es el tener un vino propio, acabara siendo un factor
preocupaciones adicionales en lugar de una satisfacción y motivo de orgullo, decidimos crear el proyecto “Thalassa Taller de Vino”. ( Posteriormente renombrado como “La Melonera” en homenaje a una de las variedades de vid recuperadas por nuestro equipo técnico).
“XXXX” combina el disponer de una gran hacienda solariega, de representación y ocio, ubicada en un paraje natural protegido, y con una oferta de actividades de ocio extensa – Más de 40 campos de Golf, hípica, deportes Náuticos, ASCARI Race Resort, etc.. – y a la vez de una hacienda bodeguera,
con su propia plantación de viñedo, en un auténtico taller de vino en permanente
evolución.

The ‘Talaza Wine Workshop’, a viticultural real estate project, known as ‘La Melonera’, began more than seven years ago thanks to the initiative of, among others, Javier Suqué Mateu (owner of Castillo Perelada) and José Luis Pérez (catalyst of the Priorato Denomination of origin, with worldwide famous wines such as L'Ermita, Clos Dofí, Clos Mogador and Mas Martinet).
The evolution of Spanish new cuisine over the last 20 years has placed four Spanish chefs in the world's top ten ranking, with Ferrán Adriá as number one since 2002. ‘La Melonera’ strives to embrace these experiences, with a philosophy of constant multidisciplinary research in viticulture, preparation and final blending of our wines.
Numerous professionals and entrepreneurs turned to José Luis Perez or Javier Suque to seek advice on how to create a wine with a personal touch, like those of the big chateaux Bordelais, Vega Sicilia or the renowned Brunello di Montalcino. However, many were forced to give up that dream due to demanding investments and the difficulty in creating something truly unique, combined with the fear that a hobby or passion, as is creating and producing one's own wine, could be a source of additional worries rather than a satisfaction or an achievement. This is what prompted us to create the ‘Talaza Wine Workshop’, later named ‘La Melonera’, in honour of one of the varieties of vine our technical team managed to restore.
‘XXXX’ combines a splendid, leisure-filled, sunny hacienda, located in a protected natural area, with a wide range of activities -more than 40 golf courses, riding centres, water-sports facilities, the prestigious ASCARI Race Resort, etc.- and a bodega with its own vineyard plantation, in an authentic wine workshop in constant evolution.
